Specific nuclear binding of angiotensin II by rat liver and spleen nuclei.

چکیده
1. Nuclei were isolated from rat liver and spleen by a standard technique which preserved the nuclear membrane. Electron microscopy, DNA/protein ratios and S'-nucleotidase determinations codfirmed the purity of the preparation. 2. Both liver and spleen nuclei specifically bound '251-labelled angiotensin I1 (ANG 11) with high affinity (Kd Z 1-0 nmol/l). Saralasin ANG I11 and ANG I1 competed with tracer for ligand whereas ANG I did so less effectively and neurotensin not at all. 3. These results suggest the presence of specific nuclear intracellular ANG I1 receptors and raise important questions regarding the mechanism of action of this peptide.

Nuclear angiotensin receptors induce transcription of renin and angiotensinogen mRNA.
The observation that nuclei from hepatic tissue exhibit specific angiotensin II (Ang II) binding led us to explore whether Ang II modulates mRNA in general, mRNA specific for renin system components, or both. Nuclei from hepatic tissue exhibited a single high-affinity (Kd = 0.4 nmol/L) Ang II-specific binding site, which was associated with increased RNA transcription. Whereas total RNA extract...
متن کاملRadiosynthesis and evaluation of ytterbium-175 labeled bleomycin as therepeutic agent
Introduction: Bleomycins are DNA-binding biomolecules, which can be used as targeted therapy carriers when labeled with particle-emitters such as Yb-175. In this work the development of Yb-175 bleomycin (175Yb-BLM) has been reported. Methods: Yb-175 chloride was obtained by thermal neutron irradiation (3 × 1013 n.cm-2.s-1) of natural Yb2O<...
متن کاملA liver-specific factor and nuclear factor I bind to the rat alpha-fetoprotein promoter.
Proteins putatively involved in the transcriptional control of rat (alpha-fetoprotein (AFP) gene expression in the liver were identified by analyzing the in vitro binding of proteins from nuclear extracts of fetal and adult rat livers to the cloned rat AFP promoter region (-197 to +48) using a combination of gel shift and DNase I footprinting assays. Three stable and specific high affinity comp...
متن کاملStudies on the nuclear binding of steroid hormone-receptor complex; binding of liver and thymus dexamethasone-receptor complex and prostate dihydrotestosterone-receptor complex to nuclei from various tissues.
To examine the binding specificity of steroid hormone-cytoplasmic receptor complexes to nuclei, binding of 3H-dexamethasone (Dex)-liver, 3H-Dex-thymus and 3H-dihydrotestosterone (DHT)-prostate receptor complexes to nuclei from liver, prostate, thymus, spleen and kidney was studied. It was observed that a significant amount of steroid-receptor complexes was bound to any nuclei used in the presen...
متن کاملInteraction of intracerebroventricular injection of captopril and histamine on water intake in adult male Wistar rats
Several evidences have indicated that many nuclei in the brain have critical role in water intake. Angiotensin II is one of the most important neurotransmitters involved in regulation of fluid homeostasis. In this study, the interaction of i.c.v. injection of captopril and histamine on water intake in adult male water-deprived rats was investigated. Rats were implanted with guide cannula aimed ...
